No reply generally means that they aren't interested. But it can also mean that they haven't both looked at your message and profile yet. But if you keep messaging, it can often turn "one of us hasn't looked at the profile yet" into "not interested". This is especially true for when SMs send messages, but it applies to couples as well.
Probably best to send a short message expressing interest, two to four sentences referring to something you found attractive in their profile, and send. After that, the ball is in their court. Just keep in mind that making connections strictly online is usually a low success method. Going to parties or clubs is going to pay off more often.
